    PhD in cellular and molecular biology

    Mes travaux de thèse me permettant d'accéder au titre de docteur en biologie cellulaire et moléculaire ont porté sur la mise au point et la caractérisation d'un modèle de cellules persistantes au traitement de chimiothérapie dans le cas du cancer du sein triple négatif. Pour cela j'ai effectuer une revue de la littérature et discutée avec des cliniciens pour comprendre le schéma de traitement clinique et comment l'adapter in vitro. Once established, le modèle a été caractériser à l'aide de technique sans a priori (proteomics, metabolomics) and the clinical relevance of potential targets identified was searched in public databases (ex: cbioportal). Le rôle des cibles identifiées a été validé dans ce même modèle de façon fonctionnelle par notamment l'analyse du métabolisme énergétique (Seahorse) and an in vivo xenograft assay. L'ensemble de ces travaux ont mené à la publication en 1ère auteur de 2 scientific articles in international journals (Winter et al, cancers, 2021; Winter et al, Neoplasia, 2023) ainsi qu'à la publication en co-auteure d'un 3ème articles (currently being submitted).

    My post-doc project focused on modeling persistence to chemotherapy treatment. I was able to develop a model of treatment persistence in 2D and 3D culture on conventional culture cells as well as cells from PDX. The 2D culture model was characterized at the molecular level by CRISPR-screen and RNA-seq as well as changes in mitochondrial metabolism. (Seahorse). The treatment sensitivity of this model was tested by analyzing drug libraries and carrying out their apoptotic profile..
